Determination of β-Sitosterol in Pygeum Bark Extract by HPLC
1. Standard solution preparation
Weigh accurately a β-Sitosterol 15.0mg standard (Sigma company the U.S.A.) in a 25ml volumetric flask， and add a solution of Methanol to mark.
2. Sample solution preparation
Weigh accurately 100mg sample in 25ml volumetric flask, then add 20ml solution of Methanol to mark and preparation.
3. Test conditions:
Column: C8 (150 mm×4.6 mm, 5μm)
Mobile phase: Methanol : 0.1% H3PO4 Solution (98∶2, v/v)
Flow rate: 1.0 mL/min,
Detection wavelength: 210 nm
Column Temperature: 25 ℃.
4. Process
Separately inject equal volumes (about 20 uL) of the Standard solution and the Sample solution into the chromatograph, record the chromatograms, and measure the responses for the major peaks.
